Studies of the rhizoplast from Naegleria gruberi.
A procedure utilizing homogenization and centrifugation in a low ionic strength buffer containing Triton X-100, has been used to facilitate the isolation of the rhizoplast from flagellates of Naegleria gruberi. This has enabled a study to be made of the physical and biochemical properties of this organelle. The rhizoplast is shown to be a proteinaceous structure with chemical properties similar...
متن کاملSmall-subunit ribosomal RNA sequence from Naegleria gruberi supports the polyphyletic origin of amoebas.
We have sequenced the small-subunit ribosomal RNA gene of the amoebo-flagellate protozoan Naegleria gruberi. Comparison of this sequence with the rRNA sequences of other eukaryotes resulted in a phylogenetic tree that supports the suggested polyphyletic origin of amoebas and suggests a flagellate ancestry for Naegleria.
متن کاملMolecular cloning of, and phylogenetic analysis of, an actin in Naegleria fowleri.
We cloned and sequenced an intronless actin gene from the amoebo-flagellate Naegleria fowleri, LEE strain, an opportunistic pathogen of man. Codon usage and third-position-codon nucleotide frequency were significantly different from Acanthamoeba, another amoeba genus which also includes opportunistic pathogens of man. Between the two amoebae, actin peptide sequences were 92.8% similar, while nu...
متن کاملDescription of a New Species with a Remarkable Cyst Structure in the Genus Naegleria: Naegleria angularis sp. n
A vahlkampfiid amoeba was isolated from a water sample collected in Peru. Ribosomal DNA sequence analysis showed that the isolate belongs to the genus Naegleria, that it represents a novel species and that its closest relative is N. pussardi. Although there is very little sequence difference between the new isolate and N. pussardi, and both species grow at 40°C, their cyst morphology is very di...
متن کاملAncestral centriole and flagella proteins identified by analysis of Naegleria differentiation.
Naegleria gruberi is a single-celled eukaryote best known for its remarkable ability to form an entire microtubule cytoskeleton de novo during its metamorphosis from an amoeba into a flagellate, including basal bodies (equivalent to centrioles), flagella and a cytoplasmic microtubule array. Our publicly available full-genome transcriptional analysis, performed at 20-minute intervals throughout ...
